     THESIS
    Early detection of colorectal adenocarcinoma using antibodies against minichromosome maintenance (MCM) proteins.

    This translational work was carried out as a collaborative venture between the Medical Research Council Cancer Cell Unit, Hutchison/MRC Research Centre, Cambridge and the Colorectal Unit, Department of General Surgery, Addenbrooke's Hospital, Cambridge.

    I aimed to develop a novel, non-invasive method for the detection of colorectal cancer in stool. I initially designed a cell line model to optimise retrieval of neoplastic colonocytes from the stool surface and subsequent MCM protein detection. Cell yield was significantly increased if the stool was kept on ice, and addition of a mucolytic agent to the suspending solution significantly increased cell yield.

    These findings were taken forward into a clinical evaluation study, which had ethical approval. I recruited 52 subjects with colorectal cancer and 35 control subjects. Using three different techniques for colonocyte retrieval, MCM-positive cells, identified by immunocytochemistry, were retrieved from the stool of 49 of 52 subjects with colorectal cancer and none of 35 controls.

    In an attempt to identify potential sources of false-positive results, I carried out an immunohistochemical assessment of MCM protein expression in inflammatory bowel disease. This demonstrated aberrant expression of MCMs in the superficial aspect of colonic glands in active ulcerative colitis and active Crohn's disease.

    The work identified a novel, stool-based method for colorectal cancer detection, and resulted in the award of a Master of Surgery degree from the University of Cambridge, peer-review publications (including first author papers in high impact factor journals such as the Lancet and Nature Reviews Cancer) and several prizes and awards (including the Patey Prize and John of Arderne Medal). It also highlighted a potential method for stool-based monitoring of inflammatory bowel disease activity, and provides avenues for future research in these areas. Further funding was subsequently awarded to allow the appointment of a research fellow to continue with this work.

     INTERESTS & ACHIEVEMENTS
    My main interests have always been sporting, particularly rugby union. I won my Cambridge Blue in 1994, playing scrum-half for the winning Cambridge University side in the Varsity Match at Twickenham. I made over 50 appearances for The Blues, and won LX Club Colours and Under 21 Colours, captaining all sides including The Blues. At school, I represented England Schools 18 Group.
